What the fuck do you do if you break down on the "out in the sticks" part of it!?. Can be days without a car passing I'd say. Plus, wouldn't be the easiest to describe where you are over the phone.
Hours, yes.   Days, is a bit of an exageration.

It's out in the sticks. Not outer-Mongolia.

Anyone know if New York-New Orleans (stopping off along the way) is practical in a 2-3 week period?

And if so which route or mode of transport would work best?

The one thing I will say is, if you're young with no ties just do it. I never did and probably never will, but it's something I'd like to do. Trouble is, her indoors prefers Greece.. so fairly fucked on that one...   :butt

And if so which route or mode of transport would work best?

Used to drive over the road .. that's a 1 day run, non-stop.  I'd take I-95 down the eastern seaboard (see some cool sights- Washington, Philly, N & S Carolina) then cut across from Jacksonville, FL, or Orlando (Disney World?) on I-10 to NO.
OR, cut across to I-81 & down through VA, TN, (Mountains) then I-40 across through Nashville to Memphis, and I-55 down to NO.
Either way, you'll see some great sights. If you wanted you could round trip it ... down one way and back up the other very easily. Figure on 4-5 days driving 4-5 hours for each leg. I'd do convertible if in the summer .. ride w/ A/C or Air, whichever you like. ;) 
If you really wanted to branch out, in that amount of time you could easily swing by Chicago and down, or even go clear to Cali with little to no effort driving wise.
Advice for any road trip in US - book hotel/motel w/ a pool !! Nice to stretch/ease after driving so much.

After watching the Billy Connolly programme tonight, it's confirmed to me that I want to do a road trip across a route of America.

Very jealous of the people in here that have done so already.

just got the second part of it, have the rest on series link though. Pretty good show, particularly enjoyed him in Abe Lincoln's house

This is where it pays to pick up that hitcher bloke with the 1000 yard stare.  He can then introduce you to some friends of his in the nearest little town which is not found on any map.

Cheers for that very detailed answer! Yeah thinking a round trip would work best. Would absolutely love to do it, just desperate to visit both New York (again) and New Orleans (first time). Will be suggesting this to my travelling buddy.

2-3 Weeks? American Airlines Flight 543 from JFK takes 3 hours and 20 minutes.
